Sujet de Recherche :
Les substrats énergétiques utilisés par les procaryotes sont extrêmement divers et comprennent même des substances toxiques pour la plupart des organismes. L’arsenic apparait ainsi de plus en plus clairement comme un substrat microbien bioénergétique majeur, utilisé dès l’archéen. Notre objectif de recherche est non seulement de comprendre la place du métabolisme de l’Arsenic dans l’évolution globale des processus énergétiques et de la vie sur terre, mais également de décortiquer les mystères encore nombreux des mécanismes moléculaires du fonctionnement des enzymes impliquées dans le métabolisme énergétique de l’Arsenic : arsénite oxydase Aio, arsénite oxydase alternative Arx et arséniate reductase Arr. Ces questions sont abordées selon la spécificité de notre équipe BIP09, c’est-à-dire l’approche phylogénétique conjointe à l’expérimentation biochimique (enzymologie, purifications), biophysique (spectroscopie optique et RPE) et microbiologique. Celles-ci nous amènent à étudier 1) les déterminants moléculaires des propriétés d’autres enzymes à Molybdène (Mo) que celles impliquées dans le métabolisme de l’Arsenic, comme la TMAO réductase TorA, la Sulfite oxydase Soe 2) la phylogénie d’enzymes autres que celles du métabolisme de l’Arsenic, en lien avec celui-ci, par exemple les enzymes du métabolisme du soufre ou du nitrate.
